Refactoring Gene Clusters for Yeast Heterologous Expression

In the example below, we refactor one pathway with multiple designs.
This pathway has four genes:




Version 0.x

In Version 0.x, each gene is driven by the ADH2 promoter, with ADH2 terminator.

In Version 0.1, each gene is driven by the ADH2 promoter, with ADH2 terminator in pRS series 2µ plasmids.

Version 1.x

In Version 1.x, genes are driven by strong constitutive promoters.

In Version 1.1, genes are driven by TEF1p, FBA1p and PDC1p followed by PGI1t, ENO2t, and TDH2t, respectively. All constructs are in pRS series 2µ plasmids.

In Version 1.2, genes are driven by TEF1p, FBA1p and PDC1p, followed by PGI1t, ENO2t, and TDH2t, respectively. All constructs are in pRS series CEN ARS plasmids.
